In order to synthesize proteins locally, messenger RNAs (mRNAs) and components of translational machinery must be transported to neurites. This unconventional view of protein distribution to different neuronal compartments has been finally accepted by the scientific community. However, in the 19th century, the possibility of neurites, especially axons, producing their own proteins locally was already hypothesized (review in Bolton, 1901). It was classically thought that proteins that support dendritic and axonal functions are synthesized in the soma and then transported to the target compartment at peripheral sites of the neuron. The origin of neuritic proteins (both dendritic and axonal) has been discussed for years. These implies that compartmentalized signaling events are required and therefore neuronal proteins must be asymmetrically distributed. To maintain a proper function, each neuronal compartment needs to react temporally and spatially in an acute manner in order to rapidly adapt to changes in the environment. Whereas dendrites receive signals, the cell body processes them and axons are responsible for transmitting information to adjacent neurons. This extremely polarized morphology reflects the also polarized function of neurons. We have observed that Aβ increases the synthesis of neuritic proteins as well as the fraction of translating RNAs in distal sites of the neurite, suggesting an induction of local protein synthesis. Aβ is involved in the pathology of Alzheimer’s disease and was previously reported to induce local translation in axons and dendrites which in turn contributes to the disease. To test the validity of our analyses we have compared control neurons to Aβ 1 – 42-treated neurons. Colocalization between RNA and protein in neurites correspond to newly synthesized proteins arising from localized RNAs and represent localized translation sites. Objects corresponding to RNA or protein and colocalized objects (RNA and protein) are quantified along individual neurites. Resulting pixels are converted into objects and selected by automatic masking followed by signal smoothing. After imaging, signals corresponding to neuritic RNAs and proteins are filtered with a Laplacian operator to enhance the edges. Newly synthesized proteins are tagged with puromycin and endogenous RNAs labeled with SYTO. Here we describe an object-based analysis used to visualize and quantify local RNA translation sites in neurites. Thus local translation events can be easily overlooked under the microscope. For instance, in hippocampal neurons levels of newly synthesized somatic proteins can be more than 20–30 times greater than translation levels of neuritic proteins. The relevance of local protein synthesis in neuron development, maintenance and disease has not been fully acknowledged until recent years, partly due to the limited amount of locally produced proteins. Strength of a material is obviously important. ![]() Examples of code-prescribed deflection limits and live load values are:īedrooms and habitable attic floors L/360 & 30 psfĪttic floors with limited storage L/240 & 10 psf. These limits are based on live loads and activities experienced in specific rooms of a house. Typical deflection limits referenced in code books are L/360, L/240 or L/180. Drywall attached to the underside of this system is not expected to crack when the floor joist system deflects 1/3″. For example: a floor joist appropriately selected to span 10 feet with an L/360 limit will deflect no more than 120″/360 = 1/3 inches under maximum design loads. They are expressed as a fraction clear span in inches (L) over a given number. Maximum deflection limits are set by building codes. Only live loads are used to calculate design values for stiffness. In other words, how much a joist or rafter bends under the maximum expected load. Stiffness of structural members is limited by maximum allowable deflection. Perhaps the joists were strong enough if they didn’t break! But lack of stiffness leads to costly problems. For example, first-floor ceiling plaster would crack as occupants walked across a second-floor bedroom that was framed with bouncy floor joists. Strength and stiffness are equally important. Beams, studs, joists and rafters act as a structural skeleton and must be strong enough and stiff enough to resist these loads. The house acts as a structural system resisting dead loads (weight of materials), live loads (weights imposed by use and occupancy), like snow loads and wind loads. This article will focus on how simple beams like joists and rafters react to loading. If, when the loads of the house are combined, the house weighs more than the soil can support – the house will sink until it reaches a point at which the soil can support the load. Remember when your science teacher said: every action has an opposite and equal reaction? Well every building load has an equal “reaction load”. The structural goal of a house is to safely transfer building loads (weights) through the foundation to the supporting soil. A complete analysis of wood’s mechanical properties is complex, but understanding a few basics of lumber strength will allow you to size joists and rafters with the use of span tables. I'm kinda surprised how mediocre the animation was, especially in an action series for BONES, but they were probably dumping all their resources into Darker than Black at the time. I can see why some people like it, mainly due to the atmosphere, but I'm left apathetic to the anime as a whole. It's not a good thing when the ending of the show reminded me of Blood-C. I wasn't impressed with the characters, art direction, animation, storyboarding, or general story premise. It's one of BONES more forgotten shows and I can see why. Duckroll and somebody else here recommended it so I decided to give it a whirl. I finally got around to watching this show in the past few weeks as I've been meaning to start it for a few months now. IIRC the manga (no spoilers, just impressions) The series has some crazy good action scenes, and the titans are awesome villains. I'm not sure how it could have been better integrated though - maybe after the fight? Also, the flashback occurs in a weird spot, kind of interrupting the "wdf" aspect of the twist. Two things bugged me about the most recent reveal, in hindsight - the rest of the troop keeps walking long after Mikasa stops, and this looks a little awkward, especially because they do eventually stop but by that point should be too far to hear. They just didn't seem super important, but they were there, and their prior scenes all make sense with this new context. When I read the reveal in the manga, I was like "who the fuck are all these characters and why should I care?" but all of them - Ymir, Historia, Reiner, Bert - they've all been there from the beginning. ![]() Going through the series again with the knowledge of the most recent reveal was super interesting. The four year gap between seasons somehow imprinted on my brain that there was a time skip between them of a few weeks, but nope - the most recent episode happens just a couple days after the Female Titan incident. Should have done it before the second season started though, not after. Really enjoyed going through the show again, really reignited my love of the series. So I'm not going to entertain those kinds of episodes with my enthusiasm anymore. It's like the anime industry is continuously whispering in my ear: "Original Super Robots are dead, that's your lot." ![]() Not some random episode of a show with a plot that has nothing to do with super robots. I want to see 2D super robot animation from a full-length 2D Super Robot show. It's honestly painful at this point, seeing great 2D Super Robot animation WASTED on one-off forgettable tributes to a subgenre of mecha from a bygone age. ![]() This exact same thing happened with Flip Flappers episode 8 and one particular episode of Active Raid that I can't be bothered to remember the number. I'd get all excited over some good 2D robot sakuga and jump up and down about how good it feels to see a good mecha thing again and we'd all forget about it like it was nothing of significance. Normally that'd have me racing to watch the episode but then it hit me like a ton of bricks to my face on how it was all going to go down. You know! The one with the cool mecha on it! The area of Thunderbird where you activate Cards View is called the Message List Header. More features such as a message preview line and sender avatars will be added in the future.
